Clinical evaluation of Zhenqi Fuzheng granules combined with trastuzumab and pertuzumab in treatment for HER2-positive breast cancer

Abstract：Objective To evaluate the clinical efficacy of Zhenqi Fuzheng granules combined with trastuzumab and pertuzumab in treatment for human epidermal growth factor receptor 2(HER2)-positive breast cancer.Methods A total of 78 HER2-positive breast cancer patients who were diagnosed and treated in the First Affiliated Hospital of Henan University of Science&Technology between January 2022 and January 2025 were selected and randomly divided into the combination group(n=39)and the dual-target group(n=39).The dual-target group received intravenous administration of trastuzumab and pertuzumab once every three weeks per treatment cycle,for a total of four cycles.The combination group additionally received oral Zhenqi Fuzheng granules from day 1 of the first cycle until the end of the fourth cycle.The clinical efficacy and the incidence of adverse reactions were compared be-tween the two groups.The Functional Assessment of Cancer Therapy-Breast(FACT-B)scores and T-lymphocyte subset levels before treatment and after four cycles were compared between the two groups.Results The objective response rate(ORR)was higher in the combination group than that in the dual-target group(89.74%(35/39)vs 71.79%(28/39),χ2=4.044,P<0.05).After four cycles,the physiological status,functional status,emotional status,social/family status and additional breast concern of FACT-B subscales,the total FACT-B score,as well as the CD4+percentage and the CD4+/CD8+ratio were higher in the combination group((20.58±2.30)points,(18.95±2.41)points,(15.01±1.37)points,(19.87±1.38)points,(24.73±2.61)points,(99.14±8.62)points,(51.21±6.38)%,and(2.00±0.51),respectively)than those in the dual-target group((16.73±2.14)points,(16.42±1.69)points,(13.84±1.31)points,(17.91±1.42)points,(22.46±2.19)points,(87.36±7.43)points,(46.52±6.17)%,and(1.42±0.47),respectively)(t=7.653,5.368,3.855,6.182,4.161,6.464,3.300,5.223,all P<0.05).The CD8+percentage in the combination group was lower than that in the dual-tar-get group((25.64±3.27)%vs(32.71±3.58)%;t=9.106,P<0.05).There was no statistically significant difference in the incidence of adverse reactions between the two groups(12.82%(5/39)vs 23.08%(9/39);χ2=1.393,P=0.238).Conclusion Zhenqi Fuzheng granules combined with trastuzumab and pertuzumab can effectively regulate the immune function,improve the quality of life and demonstrate significant clinical efficacy in HER2-positive breast cancer patients.
