Efficacy and safety of dual-target therapy of trastuzumab and pertuzumab combined with neoadjuvant chemotherapy in the treatment of HER2 positive breast cancer patients

Abstract:Objective To analyze the efficacy and safety of the dual-target therapy of trastuzumab and pertuzumab combined with docetaxel and carboplatin neoadjuvant chemotherapy(TCHP)regimen in the treatment of human epidermal growth factor re-ceptor-2(HER2)positive breast cancer patients.Methods The study subjects were 104 patients with HER2 positive breast cancer admitted to Yuncheng Central Hospital Affiliated to Shanxi Medical University from February 2022 to February 2025.48 patients in the control group were treated with the TCbH single-target regimen(docetaxel+carboplatin+trastuzumab),while 56 patients in the study group were treated with the TCbHP dual-target regimen(docetaxel+carboplatin+trastuzumab+pertuzumab).The clini-cal efficacy,tumor growth factors,serum tumor marker levels,and safety were compared between the two groups.Results When pathological complete response(pCR)was defined as ypT0/is ypN0,the pCR rate in the control group was lower than that in the study group(70.83%vs 87.50%,χ2=4.455,P<0.05).When pCR was defined as ypT0 ypN0,the pCR rate of the control group was lower than that of the study group(52.08%vs 73.21%,χ2=4.977,P<0.05).After treatment,the levels of vascular endothelial growth factor A(VEGF-A)and VEGF-B in the study group((76.01±6.54)ng·mL-1 and(64.64±6.81)ng·mL-1,respectively)were both lower than those in the control group((92.40±8.52)ng·mL-1 and(78.38±7.59)ng·mL-1,respectively)(t=11.084,9.729,both P<0.05).After treatment,the levels of carcinoembryonic antigen(CEA),tissue polypeptide specific antigen(TPS),and carbohydrate antigen 153(CA153)in the study group((2.04±0.51)pg·mL-1,(15.16±1.39)μg·L-1,and(22.82±9.46)U·mL-1,respectively)were all lower than those in the control group((2.94±0.72)pg·mL-1,(19.62±2.85)μg·L-1,and(29.38±8.50)U·mL-1,respectively)(t=7.431,10.366,3.693,all P<0.05).There was no statistically significant difference in the incidence of adverse reactions between the two groups(P>0.05).Conclusion The combination of dual-target therapy of trastuzumab and pertuzumab with neoadjuvant chemotherapy TCHP regi-men for HER2 positive breast cancer can effectively enhance the clinical efficacy,reduce the levels of tumor growth factors and tu-mor markers,and demonstrate good safety.
