Influencing factors for adverse reactions in breast cancer patients receiving olaparib maintenance therapy

Abstract:Objective To explore the influencing factors for adverse drug reactions(ADRs)in breast cancer patients receiving olaparib maintenance therapy so as to provide a reference for the safe clinical use of olaparib.Methods The medical records were collected from 97 patients who underwent non-breast conserving surgery for breast cancer and olaparib maintenance therapy in Nei-jiang Traditional Chinese Medicine Hospital from March 2020 to March 2024 and were retrospectively analyzed.The patients were divided into the ADR group(69 cases)and the ADR-free group(28 cases)based on the ADR classification results.The clinical data were compared between the two groups,and the influencing factors for ADRs in the breast cancer patients undergoing olaparib main-tenance therapy were explored through univariate and multivariate logistic regression analysis.Results The main ADRs in the pa-tients included anemia(54.64%),fatigue(46.39%),nausea(18.56%)and neutropenia(17.53%).The proportion of patients who had a history of discontinuation or dose reduction due to myelosuppression during platinum-containing chemotherapy,clinical stage Ⅲa and grade≥3 anemia during platinum-containing chemotherapy was higher in the ADR group(71.01%,63.77%,30.43%)than in the ADR-free group(46.43%,39.29%,10.71%)(χ2=5.220,4.863,5.974,all P<0.05).Logistic regression analysis showed that a history of discontinuation or dose reduction due to myelosuppression during platinum-containing chemotherapy,high clinical stage level and grade≥3 anemia during platinum-containing chemotherapy affected the occurrence of ADRs in patients.Conclusion A history of drug discontinuation or dose reduction due to myelosuppression during platinum-containing chemotherapy,high clinical stage level and grade≥3 anemia during platinum-containing chemotherapy are influencing factors for the occurrence of ADRs in the breast can-cer patients receiving olaparib maintenance therapy.
