Effect of different doses of growth hormones combined with leuprorelin on treatment of girls with idiopathic central precocious puberty

Abstract：Objective To observe the efficacy of different doses of growth hormones(GH)combined with leuprorelin on treat-ment of girls with idiopathic central precocious puberty(ICPP).Methods A total of 80 girls with ICPP who were treated in the Chongzhou Maternal and Child Health Hospital from January 2023 to August 2024 were prospectively enrolled in the study and ran-domly divided into the high-dose group and the low-dose group,with 40 cases in each group.The high-dose group received treatment with 0.15 IU·kg-1·d-1 of growth hormone plus leuprorelin injection,while the low-dose group received treatment with 0.10 IU·kg-1·d-1 of growth hormone plus leuprorelin injection.The growth indicators(annual growth rate,adult final height,bone age maturation rate),levels of sex hormones(luteinizing hormones,follicle-stimulating hormones,estradiol),uterine and ovarian volumes,and adverse reactions were observed and compared between the two groups.Results There was no significant difference in the total effective rate of treatment between the two groups(92.50%vs 80.00%)(χ2=2.635,P>0.05).There were no significant differences in the annual growth rate,adult final height,and bone age maturation rate between the two groups after the treatment((11.25±1.24)cm·year-1 vs(11.72±1.13)cm·year-1,(1.12±0.26)cm·year-1 vs(1.03±0.21)cm·year-1,(157.81±2.36)cm vs(158.73±2.41)cm)(t=-1.772,1.703,-1.725,all P>0.05).There were no significant differences in the levels of luteinizing hormones,follicle-stimulating hormones and es-tradiol between the two groups after the treatment((2.66±0.54)IU·L-1 vs(2.81±0.73)IU·L-1,(11.72±1.31)IU·L-1 vs(11.23±1.22)IU·L-1,(21.01±2.66)pg·mL-1 vs(19.94±2.54)pg·mL-1)(t=-1.045,1.731,1.840,all P>0.05).There were no significant differences in the uter-ine volume and ovarian volume between the two groups after the treatment((3.72±0.77)mL vs(3.62±0.83)mL,(1.13±0.25)mL vs(1.22±0.31)mL)(t=-0.237、1.430,both P>0.05).The incidence of adverse reactions of the high-dose group was higher than that of the low-dose group after the treatment(25.00%vs 7.50%)(χ2=4.501,P<0.05).Conclusion Different doses of GH combined with le-uprorelin injection have the efficacy of regulating sex hormone levels,controlling the growth and inhibiting gonadal development of the girls with ICPP.However,0.10 IU·kg-1·d-1 of GH combined with leuprorelin shows higher safety in treatment of the girls with ICPP.
