Potential prophylactic drugs and screening for calcineurin inhibitor nephrotoxicity in kidney transplant recipients

Abstract:Objective To screen candidate prophylactic agents for calcineurin inhibitor nephrotoxicity(CNIT)after kidney transplantation using public databases and bioinformatics analysis and explore pathogen-related genes and pathways.Methods Po-tential prophylactic drugs were identified based on gene expression omnibus(GEO)and the library of integrated network-based cel-lular signatures(LINCS).Differentially expressed genes were analyzed by functional enrichment analysis and pathway analysis.The STRING database and MCODE plug-in were used to construct protein interaction networks and screen key genes.Results Mito-mycin,dactinomycin,topotecan,vemurafenib,mitoxantrone,afatinib and dasatinib were screened to reverse the expression of neph-rotoxicity-related genes of calcineurin inhibitors.The module genes with the highest scores(MCODE=4.8)were SUCLG1,ACSL4,ALDH2,ECI2,ALDH3A2,and ACOX2.Conclusion This study preliminarily explores the potential drugs and targets of nephro-toxicity caused by long-term use of calcineurin inhibitors after kidney transplantation,and provides a new idea for screening drugs to resist this nephrotoxicity.
