Efficacy and safety of polyethylene glycol losenatide injection in treatment of patients with type 2 diabetes mellitus complicated with non-alcoholic fatty liver disease

Abstract:Objective To investigate the effect of polyethylene glycol losenatide injection on serum adiponectin(APN)level in patients with type 2 diabetes mellitus(T2DM)complicated with non-alcoholic fatty liver disease(NAFLD)and its clinical effi-cacy.Methods A total of 102 T2DM patients with NAFLD admitted to Nanyang Central Hospital from November 2021 to No-vember 2023 were included and divided into a control group and an observation group utilizing the methodology of random number tables,with 51 in each group.The patients in the control group were treated with metformin hydrochloride sustained release tablets,and those in the observation group were treated with polyethylene glycol losenatide injection.The indexes related to glucose me-tabolism,blood glucose fluctuation index,blood lipid index and adipokines and clinical efficacy were compared between the two groups.Results After treatment,fasting blood glucose,glycosylated hemoglobin,postprandial 2 h blood glucose levels((6.49±2.13)mmol·L-1,(6.88±1.55)%,(7.67±1.87)mmol·L-1)in the observation group were lower than those in the control group((8.31±2.07)mmol·L-1,(8.45±2.19)%,(9.18±2.56)mmol·L-1,respectively),the difference was statistically significant(t=4.376,4.179,3.401,all P<0.05).The blood glucose fluctuation amplitude,the mean absolute difference of daily blood glucose and the de-cal-ibration difference of blood glucose level in the observation group((4.20±0.96)mmol·L-1,(2.88±0.96)mmol·L-1,and(1.37±0.42)mmol·L-1,respectively)were lower than those in the control group((5.84±1.24)mmol·L-1,(4.69±1.32)mmol·L-1,(1.76±0.65)mmol·L-1),and the differences were statistically significant(t=7.468,7.919,3.599,all P<0.05).The levels of triglyceride,low-density lipoprotein cholesterol and free fatty acid in observation group((1.67±0.61)mmol·L-1,(2.31±0.97)mmol·L-1,(0.51±0.23)mmol·L-1,respectively)were lower than those in control group((2.65±0.75)mmol·L-1,(2.98±1.03)mmol·L-1,(0.90±0.22)mmol·L-1,respectively)(t=7.239,3.382,8.751,all P<0.05).Serum APN((5.29±1.59)ng·L-1)in the observation group was high-er than that in the control group((3.82±1.85)ng·L-1).Phosphatidylinositol proteoglycan 4 and asprosin((2.25±0.31)μg·L-1,(1.14±0.19)ng·mL-1,respectively)were lower than those in the control group((2.72±0.25)μg·L-1,(1.69±0.32)ng·mL-1,respectively),the statistical disparity was notable(t=4.304,8.428,10.554,all P<0.05).After treatment,the observation group exhibited a superior overall efficacy rate of 96.08%(49/51)compared to 84.31%(43/51)in the control group,with the discrepancy yielding statistical significance(χ2=3.991,P=0.046).No noteworthy divergence was observed in the frequency of adverse reactions between the obser-vation group and the control group(13.73%(7/51)and 11.76%(6/51)(χ2=0.088,P=0.767).Conclusion The polyethylene glycol losenatide injection has good efficacy and safety in the treatment of T2DM patients with NAFLD by alleviating clinical symptoms and regulating glucose and lipid metabolism.
