Efficacy of zoledronic acid combined with eldecalcitol in treatment of primary osteoporosis

Abstract:Objective To explore the efficacy of the combination of zoledronic acid with eldecalcitol in the treatment of pri-mary osteoporosis.Methods A total of 102 patients with primary osteoporosis admitted to The First Affiliated Hospital of Henan University from January 2022 to December 2022 were selected as the subjects of this study.All patients were divided into two groups using the random number table method.The patients in the control group(n=51)received zoledronic acid alone,while those in the study group(n=51)received a combination of zoledronic acid and eldecalcitol.The therapeutic effects,bone metabolism,bone den-sity,and adverse reactions were evaluated and compared between the two groups.Results The effective rate in the study group was 64.17%(33/51),which was significantly higher than that in the control group(41.18%,21/51)(χ2=5.667,P<0.05).The study group exhibited higher lumbar spine bone mineral density values at 6 months and 12 months post-treatment((0.69±0.12)g·cm-2 and(0.83±0.21)g·cm-2,respectively)and higher femoral neck bone mineral density values((0.65±0.05)g·cm-2 and(0.80±0.08)g·cm-2,respectively)compared to the control group(lumbar spine bone mineral density:(0.63±0.11)g·cm-2 and(0.72±0.15)g·cm-2;femoral neck bone mineral density:(0.59±0.07)g·cm-2 and(0.71±0.07)g·cm-2).The between-group differences were statistically significant(lumbar spine:F=17.593,P<0.001;femoral neck:F=26.973,P<0.001).Both groups showed statistically significant changes in bone mineral density over time(lumbar spine:F=28.257,P<0.001;femoral neck:F=325.787,P<0.001).The trend of femoral neck bone mineral density change over time was statistically significant between the two groups(F=14.575,P<0.001),while the trend of lum-bar spine bone mineral density change over time was not statistically significant between the two groups(F=1.430,P=0.242).At 6 and 12 months of treatment,the study group exhibited higher levels of osteocalcin((17.86±1.64)ng·mL-1 and(19.93±1.84)ng·mL-1,respectively),bone-specific alkaline phosphatase((91.62±6.23)U·L-1 and(97.86±8.62)U·L-1,respectively),and bone morphoge-netic protein-2((75.63±6.41)ng·L-1 and(83.64±6.47)ng·L-1,respectively)compared to the control group((16.53±1.42)ng·mL-1,(17.63±1.34)ng·mL-1,(84.63±6.23)U·L-1,(88.48±6.84)U·L-1,(70.53±6.34)ng·L-1,and(75.64±6.53)ng·L-1,respectively)(t=4.378,7.216,5.666,6.087,4.040,6.215,P<0.05).The overall incidence of adverse reactions was not significantly different between the two groups(29.41%(15/51))for the control group and(27.45%(14/51))for the study group(χ2=0.048,P>0.05).Conclusion The com-bination of eldecalcitol with zoledronic acid demonstrates superior therapeutic efficacy compared to the use of zoledronic acid alone,with significant increases in bone density and positive impacts on bone metabolism-related indicators.
