Efficacy and safety of denosumab combined with vitamin D3 in treatment of abnormal renal function and osteoporosis

Abstract:Objective To explore the efficacy and safety of denosumab combined with vitamin D3 in treatment of patients with abnormal renal function and osteoporosis (OP). Methods With the estimated glomerular filtration rate (eGFR)<60 mL·min-1·(1.73 m2)-1 as the inclusion criterion,103 OP patients admitted to Huzhou Central from September 2020 to April 2023 were col-lected as the research subjects. These patients were divided into two groups according to treatment regimens. The patients in the control group were treated with dessumab (n=49) while those in the experimental group (n=54) were treated with vitamin D3 based on the regimen in the control group. The treatment lasted for 12 months in both groups. The level of electrolyte elements and bone metabolism indicators,bone mineral density (BMD),clinical efficacy and incidence of adverse reactions were compared between the two groups before and after treatment. Results After treatment,the blood calcium and BALP levels in the experi-mental group were (2.64±0.29) mmol·L-1,(401.84±62.33) U·L-1,which were higher than those of (2.53±0.24) mmol·L-1,(325.61±51.96) U·L-1 in the control (t=2.085,6.704,both P<0.05). However,the serum phosphorus and β-CTX levels were lower in the ex-perimental group than in the control (1.30±0.12) mmol·L-1 vs (1.35±0.13) mmol·L-1,(0.47±0.08) ng·mL-1 vs (0.54±0.09) ng·mL-1) (t=2.030,4.179,both P<0.05). The lumbar spine L2~4 and total hip BMD levels in the experimental group ((0.84±0.15) g·cm-2,(0.94±0.17) g·cm-2) were higher than those in the control ((0.78±0.13) g·cm-2,(0.87±0.15) g·cm-2) (t=2.159,2.206,both P<0.05). The total effective rate of the experimental group was 92.59% (50/54),which was higher than that of 77.55% (38/49) in the control (x2=4.671,P<0.05). The overall incidence of adverse reactions was 12.96% (7/54) in the experimental group and 10.20% (5/49) in the control,showing no significant difference between the two groups (x2=0.190,P>0.05). Conclusion Desosumab combined with vitamin D3 has a good therapeutic effect on patients with renal dysfunction and osteoporosis. It can promote calcium absorption,reg-ulate bone metabolism,improve bone density,and has good safety.
