Study on the intervention of 5-HT level by Dingzhi Xiaowan regulating oxidation-reduction system

Abstract:Objective: To investigate the influence of Dingzhi Xiaowan, the antidepressant Chinese medicine, on 5-HT level and oxidation-reduction system in the depression animal model and cell model. Methods: The effects of different concentrations of Dingzhi Xiaowan on the levels of MDA, ROS, SOD and CAT in SH-SY5Y cells, C6 cells and the original rat hippocampal nerve cells harmed by gydrogen peroxide were observed. The model of chronic unpredictable mild stress (CUMS) was established to observe the effect of Dingzhi Xiaowan on 5-HT in the hippocampus, SOD and MDA levels in the hippocampus and serum of CUMS rats. Results: Under the effect of 100 μg·mL-1, 200 μg·mL-1and 500 μg·mL-1Dingzhi Xiaowan, the activity of SOD and CAT in SY5Y cells and the original rat hippocampal nerve cells was significantly increased while the activity of ROS and MDA was significantly decreased (P < 0.05). In C6 cells, the activity of SOD was significantly increased and the activity of ROS and MDA was significantly decreased (P<0.05). Medium and high concentrations of Dingzhi Xiaowan could increase the level of 5-HT in hippocampus and the level of SOD in hippocampus and serum, while it could decrease the level of MDA (P<0.01). Conclusion: Dingzhi Xiaowan had a strong regulation effect on oxidation-reduction system. Its antidepressant effect may be related to increasing the level of 5-HT level and regulating the oxidation-reduction system.
