Determination of voriconazole in human plasma by HPLC and its application

Abstract:Objective: To establish a simple and accurate method for the determination of voriconazole in human plasma. This method was used to monitor voriconazole blood concentration and promote the individualized medication. Methods: With ketoconazole as the internal standard, the chromatographic column was the Waters Symmetry C18 (4.6 mm×250 mm, 5 μm), the mobile phase was acetonitrile-0.01 mol·L-1 ammonium acetate solution (50 : 50). The detection wavelength was 255 nm with the flow rate of 1.0 mL·min-1, and the column temperature was 40 ℃ . The injection volume was 40 μL. Results: The concentration of voriconazole in the range of 0.1 – 20.0 μg·mL-1 (r = 0.9997) showed the good linearity. The lower limit of quantitation was 0.1 μg·mL-1. The recovery rates of low, middle and high concentrations were 96.24% – 101.61%, 89.06% – 93.55% and 83.76% –108.87%, respectively. The intra-day RSDs were less than 9.44% and the inter-day RSDs were less than 5.56%. The concentration of voriconazole in patient plasma were 6.36, 4.98, 0.54 μg·mL-1, respectively by the method. Conclusion: The method was sensitive, simple, accurate and applicable for voriconazole blood concentration monitoring.
