Determination of vitamin E content in the oil from seed of Hippophae rhamnoides by CGC

Abstract:Objective: To analyze the fatty acids composition in the oil from seed of Hippophae rhamnoides, and establish a capillary gas chromatography (CGC) method to determine the content of vitamin E in the seed oil of Hippophae rhamnoides. Methods: Oil was extracted from Hippophae rhamnoides seed by heated reflux solvent. GC-MS was adopted to determine its chemical constituents. The quantification of the vitamin E (Vit E) content was conducted by GC-2010 plus. Dotriacontane was acted as the internal standard. Results: Totally 28 chemical constituents were extracted. The GC system consisted of a Rtx-5 capillary column (30 m × 0.25 mm, 0.25 μm). Column temperature was temperature programming including 150 ℃ for 13 min, rose to 300 ℃by the rate of 10 ℃·min-1 and maintained 300 ℃ for 8 min. The temperature of FID detector and the injection was 300 ℃ and 310 ℃respectively. The splitless injection was adopted, Vitamin E was separated satisfactorily from dotriacontane. The calibration curve for Vitamin E was linear in the range of 0.02 – 0.43 mg·mL-1 (r = 0.9996). Conclusion: The capillary GC method was sensitive and reliable. It could be used to determine the content of vitamin E in the Seed oil of Hippophae rhamnoides.
