Determination of all-trans Vitamin A acetate in cod liver oil ointment by HPLC

Abstract:Objective: To establish the HPLC method for determination of all-trans Vitamin A acetate in cod liver oil ointment. Methods: The Vitamin A acetate in ointment was extracted by ultrasonic method, and the content of all-trans Vitamin A acetate was determinated by HPLC method. HPLC was carried out on a column of Platisil ODS C18 (250 mm × 4.6 mm, 5 μm) with mobile phase consisting of methanol-water (98 : 2). The flow rate was 1.0 mL·min-1. The detection wavelength was set at 328 nm. The column temperature was 30 ℃.Results: Vitamin A showed a good linear relationship in the range of 0.5879 – 7.3485 U·mL-1 (r =0.9999). The average recovery was 99.78% (RSD = 1.57%). Conclusion: The method is simple, accurate and repeatable, which is suitable for the content determination of all-trans Vitamin A acetate in cod liver oil ointment.
