Study on the role of clinical pharmacists for determination of the concentration of lamotrigine and oxcarbazepine metabolite in serum and individual administration

Abstract:Objective:To establish the high performance liquid chromatography (HPLC) method for simultaneous determination the concentration of lamotrigine and oxcarbazepine metabolite in serum, in order to provide individualized drug administration by monitoring drug concentration.Methods: Lamotrigine and oxcarbazepine metabolite were used as internal standard for each other. The chromatographic column was Hypersil BDS C18 column (250 mm × 4.6 mm, 5 μm). The mobile phase was consisted of methanol-water (45 : 55). The lfow rate was 1 mL·min-1. The detecting wavelength was 225 nm using diode array detector. The column temperature was 25℃. The injection volume was 10 μL.Results: As for lamotrigine, the average method recovery rates of three concentrations (0.80, 8.00, 40.00 mg·L-1) were 95.7%, 100.9% and 99.6%, respectively. As for oxcarbazepine metabolite, the average method recovery rates of three concentrations (1.12, 14.00, 56.00 mg·L-1) were 97.1%, 101.5% and 100.3%, respectively. The intra-day relative standard deviation (RSD) was less than 3%, and inter-day RSD was less than 5%.Conclusion:The HPLC method was proved to be simple and precise with a good resolution, which was suitable for the therapeutic drug monitoring (TDM) and pharmacokinetic study of lamotrigine and oxcarbazepine metabolite.
