Determination of vancomycin concentration in human plasma by HPLC

Abstract:[AbstrAct]Objective: To establish the method for determination of vancomycin in human plasma by HPLC, and investigate the effects of different precipitants to vancomycin extraction rate.Methods: Norvancomycin was used as the internal standard, the liquid chromatography was performed on Wondasil? C18-WR column (4.6 mm × 150 mm, 5 μm), the lfow rate was 1.0 mL·min-1, and UV detector wavelength was 236 nm. Using two mobile phases of methanol-0.05 mol·L-1 potassium dihydrogen phosphate buffer (pH 3.2) and acetonitrile-0.05 mol·L-1 potassium dihydrogen phosphate buffer (pH 3.2) to select the best ratio of mobile phase. The concentration and volume of protein precipitants were optimized through experiments, ifve kinds of precipitant were used to investigate the affect of plasma sample extraction rate.results: The results showed that mobile phase consisted of methanol-0.05 mol·L-1 potassium dihydrogen phosphate buffer (pH 3.2) (16 : 84) and acetonitrile-6% perchloric acid (1 : 6) as a precipitant, the extraction rate was the best.conclusion:The method for the determination of plasma vancomycin concentration was optimized and proved to be high extraction rate, reproducible, simple and sensitive, it is suitable for the determination of vancomycin concentration.
