Effect of salvianolate on pharmacokinetics of atorvastatin calcium in rats with myocardial ischemia

Abstract:Objective: To study the effect of salvianolate on the pharmacokinetics of atorvastatin calcium in rats with myocardial ischemia.Methods:The rat models of myocardial ischemia were established by injecting pituitrin into the peritoneal cavity of SD rats. Parallel control method was applied in this study. Ten model rats were evenly randomized into two groups:atorvastatin calcium (30 mg·kg-1) group and drug combination group (atorvastatin calcium 30 mg·kg-1 plus salvianolate 200 mg·kg-1). Blood samples (1 mL) were collected from the caudal vein at 0.083, 0.17, 0.25, 0.5, 1, 2, 4, 8, 12, 24, 36 h after drug administration. LC-MS/MS was used to determine the plasma concentrations of atorvastatin calcium. The pharmacokinetic parameters of the two groups were compared, and statistical analysis was performed by DAS 3.0 software. Results:When combined with salvianolate, the AUC0-t and AUC0-∞of atorvastatin calcium were increased obviously (P<0.05). The other pharmacokinetic parameters had no signiifcant changes (P>0.05). Conclusion: In the state of myocardial ischemia, salvianolate could apparently increase the bioavailability of atorvastatin calcium, while it almost had no effect on drug elimination process.
