Simultaneous determination of puerarin and paeoniflorin in Jitongning capsule by HPLC

Abstract：Objective:To establish the HPLC method for the simultaneous determination of puerarin and paeoniflorin in Jitongning capsule. Methods:HPLC was carried out on a Diamonsil C18 column (250 mm × 4.6 mm, 5μm), the mobile phase consisted of acetonitrile-0.5%phosphoric acid solution (13.5∶86.5) at the flow rate of 1.0 mL· min-1, the detection wavelength was 230 nm, and the column temperature was 35℃. Results:The linear range of puerarin was 88.512–885.120μg· mL-1 (r=0.999 8). The average recovery was 99.90%with RSD of 1.28%(n=6). The linear range of paeoniflorin was 49.28–492.80μg· mL-1 (r=0.999 9), the average recovery was 99.42%with RSD of 1.02%(n=6). Conclusion:This method is simple and accurate, with good specificity and repeatability, and can be used for quantitative control of Jitongning capsule.
