Causal association between adult-onset Still's disease and coronary heart disease coronary atherosclerotic heart disease:a Mendelian randomization study

Abstract:Objective To analyze the causal association between adult-onset Still's disease(AOSD)and coronary atherosclerotic heart disease(CHD)by using the Mendelian randomization(MR)method.Methods Two-sample MR analysis was performed via the genome-wide association study(GWAS)summary database,adopting the inverse variance weighting(IVW)method,weighted median method and MR-Egger regression method.The credibility of the results was further evaluated by the multivariable Mendelian randomization(MVMR)method.Results A causal association was identified between AOSD and CHD increased risk(IVW:OR=1.039,95%CI:1.014~1.064,P=0.001).In the MR analysis,no horizontal pleiotropy or heterogeneity was found in the instrumental variables.Sensitivity analysis using leave-one-out method(LOO)confirmed the robustness of the MR results.Additionally,after adjusting for confounding factors by MVMR method,the above causal association remained statistically significant(OR=1.104,95%CI:1.022~1.192,P=0.012).Conclusion The results of MR analysis indicate that AOSD has a causal association with CHD elevated risk.
