Study on the relationship between serum silent information regulator factor 2 related enzyme 1 and transforming growth factor-β1 expression and ventricular remodeling in patients with chronic heart failure

Abstract:Objective To investigate the relationship between the expression levels of serum silent information regulator factor 2 related enzyme 1(SIRT1)and transforming growth factor-β1(TGF-β1)in patients with chronic heart failure(CHF)and ventricular remodeling.Methods A total of 100 CHF patients admitted to Guizhou Medical University Affiliated Hospital from October 2021 to December 2022 were collected as the study subjects(CHF group),and another 100 individuals undergoing health examinations at the same hospital were collected as the control group.According to the NYHA classification,100 CHF patients were graded,including 42 cases in grade IⅡ,33 cases in grade Ⅲ,and 25 cases in grade Ⅳ.Enzyme linked immunosorbent assay(ELISA)was applied to measure serum levels of SIRT1 and TGF-β1;color Doppler echocardiography was applied to measure ventricular remodeling related indicators in CHF patients;the serum levels of SIRT1 and TGF-β1 were compared between the CHF group and the control group,and patients with different New York Heart Association Cardiac Function(NYHA)grade;Pearson correlation was applied to analyze the correlation between serum SIRT1 and TGF-β1 levels,their relationship with ventricular remodeling related indicators,and the relationship between serum SIRT1 and TGF-β1.Results Compared with controls,serum SIRT1 was decreased and TGF-β1 increased in CHF patients(P<0.05).Versus grade Ⅱ,patients in grades Ⅲ and Ⅳ had higher LV end-diastolic diameter(LVEDD),left atrial diameter(LAD),left ventricular mass index(LVMI),interventricular septal thickness(IVST),and serum TGF-β1,and lower LVEF and serum SIRT1(P<0.05).GradeⅣ patients showed further increases in LVEDD,LAD,LVMI,IVST,and TGF-β1,and decreases in LVEF and SIRT1 versus grade Ⅲ(P<0.05).Pearson analysis demonstrated that SIRT1 correlated negatively with LVEDD,LAD,LVMI,and IVST(r=-0.514,r=-0.493,r=-0.455,r=-0.462,P<0.01)and positively with LVEF(r=0.422,P<0.01).TGF-β1 correlated positively with LVEDD,LAD,LVMI,and IVST(r=0.459,r=0.617,r=0.416,r=0.447,P<0.01)and negatively with LVEF(r=-0.455,P<0.01).Serum SIRT1 and TGF-β1 were negatively correlated(r=-0.416,P<0.001).Conclusion Serum SIRT1 was significantly decreased and TGF-β1 significantly increased in patients with CHF,and both were closely related to ventricular remodeling in patients with CHF.
