Causal association between sleep traits and hypertension:a two-sample mendelian randomization study

Abstract:Objective To use two sample Mendelian randomization(MR)to analyze the causal relationship between sleep characteristics and hypertension.Methods Two sample MR analysis was conducted,with exposure factors including long sleep duration,short sleep duration and insomnia.Hypertension was the outcome.Causal effects were estimated primarily by the inverse variance weighting(IVW)method,supplemented by MR-Egger regression method,weighted median method,and weighted mode method,with IVW as the main analysis method.To enhance robustness,sensitivity analysis was conducted using Cochran's Q test,intercept obtained from MR Egger regression,funnel plot,and leave-one-out method.Results The IVW analysis showed that short sleep duration was associated with increased hypertension risk(OR=1.152,95%CI:1.091~1.202,P=0.004)and insomnia was associated with increased risk(OR=1.742,95%CI:0.089~0.619,P=0.003).No significant causal effect was found for long sleep duration(P>0.05).Cochran's Q tests indicated no significant heterogeneity for any exposure(P>0.05).The MR-Egger intercepts and funnel plots showed no evidence of horizontal pleiotropy(P>0.05).Conclusion There is a causal relationship between short sleep time,insomnia,and hypertension,while there is no genetic predictive causal relationship between long sleep time and hypertension.
