Regulation and possible mechanisms of paraventricular nucleus of hypothalamus to ventricular stability after myocardial infarction

Abstract:Objective To prove the regulating effect of paraventricular nucleus of hypothalamus(PVN)on electrical stability of infarcted myocardium,and observe the physiological and pathological changes of PVN during myocardial infarction(MI).Methods ①Male SD rats(n=36)were selected and randomly divided into 3 groups(each n=12).Acute myocardial infarction(AMI)was induced through ligating the anterior descending coronary artery in all rats.After 24 h,PVN was stimulated by 0 mA,1.2 mA and 3 mA,respectively,in 3 groups.The incidence of ventricular arrhythmias(VA)and atrial arrhythmia were observed.②Male SD rats(n=24)were selected and divided into sham-operation group and MI group.The myocardial electrophysiological activity was detected in 2 groups,and gliacyte activation in PVN and level of interleukin-6(IL-6)were observed in 2 groups.Results ①There was typical arrhythmia occurred after stimulating PVN,VA incidence was 91.6%and incidence of atrial arrhythmias was 16.6%(P<0.05).②The threshold of ventricular fibrillation was 8.0 V in sham-operation group and 7.5V in MI group(P<0.05).③The level of IL-6 in PVN was significantly higher in MI group than that in control group(P<0.05).The activations of small gliacytes and astrocytes(Ast)in PVN were more significant in MI group than those in sham-operation group(P<0.05).Conclusion PVN is a regulation center for controlling ventricular stability after MI,and the mechanism may be related to IL-6 increase induced by gliacyte activation.
