Causal relationship between unsaturated fatty acid and coronary heart disease:A Mendelian randomization study

Abstract:Objective To analyze the causal relationship between unsaturated fatty acid(UFA)and coronary heart disease(CHD)by applying Mendelian randomization(MR)study.Methods IEU OpenGWAS database was chosen and single nucleotide polymorphism(SNP)was taken as a tool and MR study was used for analyzing the causal relationship between UFA and CHD.Results The results of analysis of inverse-variance weighted(IVW)showed that UFA was negatively correlated to CHD(OR=0.369,95%CI:0.154~0.803,P<0.01).MR-Egger regression analysis(OR=0.472,95%CI:0.247~0.752,P<0.01)and weighted median estimator(WME)analysis(OR=0.347,95%CI:0.168~0.752,P=0.019)showed similar results.Cochran's Q test shows that unsaturated fatty acids have a significant impact on the correlation strength of coronary heart disease.Funnel diagram shows that the effects of all single nucleotide polymorphisms are in the same direction,which suggests that the causal relationship between unsaturated fatty acids and coronary heart disease is less affected by potential deviation.The results of funnel plot analysis show that the distribution of all SNP was basically symmetrical,indicating that the causal relationship between UFA and CHD was less affected by potential biases.The results of leave-one-out sensitivity analysis showed that after excluding any UFA,the results of IVW analysis of the remaining UFA,along with the overall results(in red),all lied on the right side of null line,suggesting that the analysis results are reliable.Conclusion UFA are a protective factor for CHD,and dietary supplementation of UFA may slow down CHD occurrence and development.
