Abstract:Objective To discuss the causal relationship between hyperuricemia and coronary heart disease(CHD)by using Mendelian randomization(MR).Methods Based on MR,single nucleotide polymorphism(SNP)related to hyperuricemia were screened as instrumental variables(IV)from genome-wide association analysis research data.The causal relationship between hyperuricemia and CHD was determined by using inverse variance-weighted(IVW),MR-Egger regression,weighted median method and Penalised weighted median.The correlation between hyperuricemia and CHD was reviewed by using OR value and 95%CI value.SNP heterogeneity was detected by using Cochran's Q test,level pleiotropy was tested by using MR-Egger regression,and sensitivity was tested by using leave-one-out method.Results Based on 3 major assumptions of MR,there were finally 271 SNP included in MR analysis.The results of IVW used in MR causal relationship analysis showed that risk of CHD induced by hyperuricemia increased by 9.5%(OR=1.095,95%CI:1.040~1.151,P=0.001).The results of heterogeneity test showed that there was no heterogeneity in SNP(Cochran Q test,Q=57.908,P<0.001).The results of MR-Egger intercept test showed that this study had no horizontal pleiotropy(intercept value=8.264×10-4,P=0.551).The results of leave-one-out method showed that no single SNP had a significant impact on the overall results.Conclusion MR analysis has showed that hyperuricemia can increase CHD risk.
