The causal relationship between non-alcoholic fatty liver disease and coronary heart disease:Mendelian randomization study

Abstract:Objective To analyze the causal relationship between non-alcoholic fatty liver disease and coronary heart disease using mendelian randomization study.Methods Using the IEU OpenGWAS database and single nucleotide polymorphism as a tool to analyze the causal relationship between non-alcoholic fatty liver disease and coronary heart disease using Mendelian randomization.Results The inverse variance weighted analysis showed a close causal relationship between non-alcoholic fatty liver disease and coronary heart disease(OR=1.013,95%CI:1.012~1.017,P<0.01).MR Egger analysis(OR=1.019,95%CI:1.004~1.016,P<0.01)and weighted median analysis(OR=1.012,95%CI:1.006~1.014,P<0.01)also showed similar results.The results of Cochran's Q-test showed that there was no heterogeneity between MR-Egger regression(P=0.415)and IVW method(P=0.326).The funnel plot results show that the distribution of all SNPs is basically symmetrical,indicating that the causal relationship between uric acid and hypertension is less affected by potential biases.The sensitivity results of the retention method indicate the reliability of the analysis results.Conclusion Nonalcoholic fatty liver disease may be an important risk factor for coronary heart disease,and controlling non-alcoholic fatty liver disease may help with the treatment of coronary heart disease.
