Causal relationship between depression and heart failure:a study bases on two-sample Mendelian randomization

Abstract:Objective To analyze the causal relationship between depression and heart failure(HF)based on two-sample Mendelian randomization.Methods The data of genome-wide association study(GWAS)related to depression and HF were collected from IEU OpenGWAS database.Mendelian randomization was conducted with single nucleotide polymorphisms as instrumental variables,through various statistical models,including method of inverse variance-weighted(IVW),weighted median method and mode-based method,to analyze the causal relationship between depression and HF.Results The results of IVW method analysis showed that there was a close causal relationship between depression and HF(OR=1.314,95%CI:1.005~1.512,P<0.001).The results of MR-Egger analysis(OR=1.519,95%CI:1.014~1.816,P<0.001)and weighted median method analysis(OR=1.162,95%CI:1.016~1.214,P=0.015)also showed the similar outcomes.The analysis results of both simple mode and weighted mode also demonstrated consistency.The results of Cochran's Q test indicated that there was no heterogeneity in MR-Egger regression(P=0.561)and IVW method(P=0.293).The results of funnel plot analysis indicated that the causal relationship between depression and HF was less affected by bias.The results of leave-one-out method analysis demonstrated that Mendelian randomization analysis employed in this study was reliable.Conclusion There is a complex correlation between depression and HF,and depression may increase HF risk and affect patient's prognosis.
