Causal relationship between vitamin D and coronary heart disease:a Mendelian randomization study

Abstract:Objective To analyze the causal relationship between vitamin D(VitD)and coronary heart disease(CHD)by applying Mendelian randomization study.Methods IEU OpenGWAS database was chosen and Mendelian randomization study was used to analyze the causal relationship between VitD and CHD.Results The results of inverse variance weighting(IVW)analysis showed that there was a negative correlation between VitD and CHD(OR=0.365,95%CI:0.172~0.825,P<0.01),which indicated that VitD was a protective factor for CHD(P<0.01).MR-Egger regression analysis(OR=0.467,95%CI:0.314~0.864,P<0.01)and weighted median estimator(WME)analysis(OR=0.383,95%CI:0.197~0.835,P=0.016)showed similar results.The results of Cochran's Q-test showed that there was no heterogeneity between MR-Egger regression analysis(P=0.415)and IVW analysis(P=0.326).The results of funnel plot analysis showed that the distribution of all single nucleotide polymorphisms(SNP)was basically symmetry,which indicated that the causal relationship between VitD and CHD was less affected by potential biases.The results of leave-one-out sensitivity analysis indicated that the analysis results were reliable.Conclusion VitD is a protective factor for CHD,and improvement of VitD level may be conducive for CHD prevention and treatment.
