Electrophysiological records of three layers of transventricular cells in hibernating myocardium in beagles

Abstract:Objective To record differential expressions of three-layer transventricular ion flows in normal and hibernating myocardium,and further explore the relationship among IKs,APD and sudden cardiac death(SCD).Methods The potassium ion current(IKs)and action potential duration(APD)of cytomembrane in epicardium,midmyocardium and endocardium in Beagle hibernating myocardium isolated by enzymatic hydrolysis were detected by using simultaneous whole-cell patch-clip technique,and transmural dispersion of repolarization(TDR)was calculated.Results Compared with normal myocardium group(group B),IKs decreased and APD increased in three-layer transventricular cells,which were more significantly in midmyocardium in hibernating myocardium group(group A).TDR increased significantly in group A compared with group B.Conclusion In hibernating myocardium,IKs decreases significantly and APD increases,and these changes of myocardial cells show heterogeneity.These changes promote TDR,and provide electrophysiological basis for occurrence of malignant arrhythmia,which may be an independent risk factor for SCD.
