Causal effect of 91circulating inflammatory cytokines on aortic aneurysm:A Mendelian randomization study

Abstract:Objective To investigate the potential causal associations between circulating inflammatory cytokines and aortic aneurysm(AA)riskbya Mendelian randomization(MR)study.Methods By extracting data from publicly available genome-wide association study(GWAS)databases for circulating inflammatory factors and aortic aneurysm(AA),bidirectional Mendelian randomization(MR),multivariable MR,and subgroup MR analyses were performed.The inverse variance weighted(IVW)method was primarily used to analyze the causal effect between circulating inflammatory factors and the risk of AA.To verify the robustness of the results,a series of sensitivity analyses were conducted,including Cochran's Q test,MR-Egger intercept test,and leave-one-out(LOO)analysis.Results The results of the bidirectional MR and multivariable MR studies indicate that genetically predicted higher levels of circulating Interleukin-10(IL-10)and Stem Cell Factor(SCF)are causally associated with a reduced risk of AA,whereas CXC chemokine ligand 10(CXCL10)and CC chemokine ligand 13(CCL13)are causally associated with an increased risk of AA.Further subgroup MR analysis results show that IL-10 and SCF reduce the risk of abdominal aortic aneurysm(AAA),while CXCL10 and CCL13 respectively increase the risk of thoracic aortic aneurysm(TAA)and AAA.Conclusion Circulating inflammatory factors IL-10,SCF,CXCL10,and CCL13 have a causal relationship with the risk of AAA,providing potential targets for the screening of AA biomarkers and the development of therapeutic drugs.
