Protective effect of stromal interaction molecule-1 inhibitor SKF96365 on atherosclerosis

Abstract:Objective To investigate the protective effect of stromal interaction molecular 1(STIM1)inhibitor SKF96365 in atherosclerosis and its underlying mechanism.Methods Ten C57BL/6J mice were used as normal controls and 30 ApoE-/-mice were given a high-fat diet to establish an atherosclerosis(AS)model,of which 10 ApoE-/-mice were left untreated(model group);10 ApoE-/-mice were treated with atorvastatin calcium(10 mg/kg·d)continuously by gavage for 8 weeks from 16 weeks of age(statin treatment group);10 ApoE-/-mice were treated with SFK96365(10 mg/kg·d)continuously by subcutaneous injection for 8 weeks from 16 weeks of age(SKF96365 treatment group).The lipid profiles as well as pathological changes atherosclerotic plaques were observed;the expression of STIM1,pJNK,NF-κB and MMP-9 proteins were detected by western blot.Results Total cholesterol(TC),triglycerides(TG),and low-density lipoprotein cholesterol(LDL-C)were significantly lower in the SKF96365 group than in the model group(P<0.05).p-JNK,NF-κB,and MMP-9 proteins were significantly lower in the SKF96365 group than in the model group(P<0.05).Conclusion SKF96365 may increase plaque stability by attenuating oxidative stress and inflammatory damage in the arteries of ApoE-/-mice and by affecting the expression of MMP-9 protein activity.
