Inhibition of atherosclerosis by gadolinium doped nano cerium oxide in mice with ApoE gene knockout

Abstract:Objective To observe the antagonism of gadolinium doped nano cerium oxide(Ce-Gd)to formation of atherosclerosis(AS)in mice with ApoE gene knockout(ApoE-/-).Methods Ce-Gd(20%)were divided,according to different doses,into normal saline control group(NS group),25 μg/ml group(low-dose group),50 μg/ml group(mid-dose group)and 100 μg/ml group(high-dose group).The influence of Ce-Gd,after treated with lipopolysaccharide(LPS),on levels of plasma inflammatory factors were observed in ApoE-/-mice.A model of ApoE-/-mice with AS was established,and divided into NS infusion group,10 mg/kg Ce-Gd infusion group(low-dose Ce-Gd group)and 50 mg/kg Ce-Gd infusion group(high-dose Ce-Gd group)at time points of 0 d,3 d and 7 d.The area of aortic AS was observed by using oil red O staining,and statistical analysis was carried out in all groups.Results Ce-Gd can decreased levels of plasma inflammatory factors induced by LPS,and AS loading in a dose-independent manner in ApoE-/-mice.Conclusion Ce-Gd can inhibit AS formation through reducing oxidative stress level in ApoE-/-mice,which is expected to become a new anti-atherosclerosis medicinal.
