Change tendency of microRNA-208a/208b in peripheral blood in patients with acute myocardial infarction

Abstract:Objective To investigate the change tendency and early diagnostic value of serum microRNA-208a/208b (miR-208a/208b) in patients with acute myocardial infarction (AMI) within 7 d after AMI attack. Methods AMI patients (n=69, AMI group) were chosen within 4 h after AMI attack from Department of Cardiology in the Third Affiliated Hospital of Henan University of Science and Technology (Luoyang Dongfang Hospital) from July 2017 to June 2018. At the same time, the volunteers (n=30) received examination of coronary angiography (CAG) and exclusion diagnosis of coronary heart disease (CHD) were chosen into control group. The samples of cubital vein blood were collected respectively after AMI attach for 4 h, 8 h, 12 h, 24 h, 48 h, 72 h, 96 h and 7 d. The expression of serum miR-208a/208b was detected by using quantitative real-time polymerase chain reaction (qRT-PCR), and level of cardiac troponin I (cTnI) was detected by using biochemical analyzer. The diagnostic value of miR-208a/208b to AMI was determined by applying receiver operating characteristic (ROC) curve at different time points. Results The levels of serum miR-208a/208b and cTnI increased significantly in AMI group than those in control group after AMI attach for 4 h, 8 h, 12 h, 24 h, 48 h, 72 h, 96 h and 7 d (P<0.05). The change curve along with time of miR-208a/208b was similar to that of cTnI, and showed a tendency of first increase and then decrease. The level of miR208a/208b reached to peak after AMI attack for 8 h, and level of cTnI reached peak after AMI attack for 12 h. The analysis of ROC curve showed that the area under curve (AUC) was 0.892 in miR208a/208b combined diagnosis for AMI (95%CI: 0.802-0.978) and Youden’s index was 0.65 after AMI attack for 8 h, and the diagnostic efficacy at time point of 8 h was higher than that at other time points. Conclusion The levels of miR-208a and miR-208b are significantly higher in AMI patients than those in non-CHD patients, and will reach to peak value after AMI attach for 8 h. They can be taken as the early diagnostic indexes for AMI.
