Influence of ultrasound microbubble combined with stem cell transplantation on myocardial microenvironment and correlation between it and serum circulating Irisin level in rats with myocardial infarction

Abstract:Objective To investigate the influence of ultrasound microbubble combined with transplantation of bone marrow mesenchymal stem cells (MSCs) on myocardial microenvironment and correlation between it and serum circulating Irisin level in rats with myocardial infarction (MI). Methods MSCs were separated, purified and cultured to collect seed cells accorded with the transplantation by using technologies of density gradient centrifugation and adherent culture. Wistar rats (n=40) were randomly divided, after successfully establishing MI model, into 4 groups: ultrasound microbubble+MSCs group (US/MB+MSCs group), MSCs group, US/MB group and control group. After treatment for 4 weeks, the level of serum Irisin was detected by using ELISA, expressions of vascular endothelial growth factor (VEGF) and CD34 were detected by using immunohistochemical staining (IHC), and microvessel density (MVD) was counted. The expression of VEGF was detected by using Western blotting assay and ELISA. Results The expressions of CD34 and VEGF and MVD were higher in US/MB+MSCs group than those in other groups (P<0.05). The expression of Irisin was the highest in US/MB+MSCs group (P<0.05). VEGF was positively correlated to Irisin (r=0.902). Conclusion Ultrasound microbubble combined with transplantation of MSCs can stimulate angiogenesis through improving myocardial VEGF, and at the same time increase level of serum Irisin. Ultrasound microbubble combined with transplantation of MSCs is positively correlated to Irisin.
