Expression of microRNA-1 in acute myocardial infarction rats and its effect on cardiomyocyte apoptosis

Abstract:Objective To investigate the expression of microRNA-1 (miR-1) in acute myocardial infarction (AMI) rats and its effect on myocardial H9C2 cell apoptosis. Methods Thirty rats aged 8-10 weeks were randomly divided into normal control group, sham operation group and model group, with 10 rats in each group. In the model group, a rat model of acute myocardial infarction was established by thoracotomy of coronary artery. The rats in the sham-operated group were only ligated without coronary artery, and the rats in the normal control group were not treated. Serum-free culture were used to construct a model of ischemic H9C2 cells, H9C2 cells were divided into three groups:control group, negative group and inhibition group. The control group did not receive any treatment. The negative group was transfected with NC, and the inhibition group was transfected with miR-1 inhibitor. the expression of miR-1 in ischemic myocardium and H9C2 cells were detected by quantitative real-time PCR (qPCR). H9C2 cells apoptosis rate was measured by flow cytometry. Western blot was used to detect the expression of cysteine-containing aspartate proteinase 3 (Caspase-3) and activated Caspase-3 (Cleaved Caspase-3) protein. Results There was no significant difference in the expression of miR-1 between the normal control group and the sham operation group (P>0.05). Compared with the normal control group and the sham-operated group, the expression of miR-1 in the ischemic myocardium of the model group was significantly increased (P<0.05). There was no significant difference in miR-1 expression, apoptosis rate and expression of Cleaved Caspase-3 protein in H9C2 cells between the control group and the negative group (P>0.05). Compared with the control group and the negative group, the expression of miR-1 in the Inhibition group of H9C2 cells was significantly down-regulated (P<0.05), and the apoptosis rate and the level of Cleaved Caspase-3 protein in the cells were significantly decreased (P<0.05). Conclusion The expression level of miR-1 in the rat myocardial infarction model is increased, which has the effect of promoting cardiomyocyte apoptosis.
