Effect of PEAR1 gene polymorphism on aspirin antiplatelet activity in patients with coronary heart disease and its correlation analysis

Abstract:Objectives To investigate the effect and correlation analysis of PEAR1 gene polymorphism on aspirin antiplatelet activity in patients with coronary heart disease. Methods A total of 364 patients with coronary heart disease diagnosed and admitted to the Department of Cardiology, Xuchang Central Hospital from April 2016 to April 2018 were enrolled. All patients were treated with aspirin for 30 days after diagnosis, and the PEAR1 genotype frequency was analyzed. . According to the PEAR1 gene polymorphism test results, the patients were divided into wild type (GG group, n=118 cases), mutant heterozygous type (GA group, n=124 cases) and mutant homozygous type (AA group, n=122 cases). Platelet aggregation rate, platelet inhibition rate, serum cyclooxygenase (COX)-1, COX-2, thromboxane (TXA)-2 and prostaglandin (PGs) levels were measured before and after treatment in the three groups. Results There was no significant difference in the basic data between the three groups (P>0.05). There was no significant difference in platelet aggregation rate, platelet inhibition rate, serum COX-1, COX-2, TXA-2 and PGs level among the three groups before treatment (P>0.05). After treatment with aspirin, platelet inhibition rate in the three groups was significantly higher than that before treatment (P<0.05). The platelet aggregation rate, serum COX-1, COX-2, TXA-2 and PGs level in GG and GA group was significantly lower than those before treatment (P<0.05). After treatment, platelet aggregation rate, serum COX-1, COX-2, TXA-2 and PGs level in GG and GA groups was significantly lower than those in AA group (P<0.05). Conclusions The polymorphism of PEAR1 gene is closely related to aspirin resistance in patients with coronary heart disease. The GG type of PEAR1 gene is the most responsive to aspirin, and the AA type of PEAR1 gene is severely resistant to aspirin.
