Relationship among expressions of advanced glycation end-products, receptor of advanced glycation end-products and matrix metalloproteinase-9 in patients with gestational hypertension

Abstract:To study the expressions of advanced glycation end-products (AGEs), receptor of advanced glycation end-products (RAGEs) and matrix metalloproteinase-9 (MMP-9) in serum and placenta tissues and correlation between them and gestational hypertension in patients with gestational hypertension. Methods The pregnant women (n=300) received prenatal Down’s screening were chosen from Chinese PLA 421 Hospital from Jan. 2014 to Dec. 2016, and their pregnant conditions were followed up. There were 7 cases of preeclampsia (preeclampsia group) and 10 cases of gestational hypertension (gestational hypertension group). Meanwhile 16 pregnant women with normal pregnant outcomes, term birth and without any complications were chosen into control group. The expressions of AGEs, RAGEs and MMP-9 in serum and placenta tissues during pregnant metaphase and later stages were detected by using double antibody sandwich ELISA and immunohistochemistry technique. Results The levels of AGEs and RAGEs increased (P<0.05) and MMP-9 level decreased (P<0.05) in preeclampsia group compared with control group during pregnant metaphase and later stages. The levels of AGEs increased (P<0.05) and MMP-9 level decreased significantly (P<0.05) in gestational hypertension group during pregnant later stages. The levels of RAGEs increased significantly (P<0.05) and MMP-9 level decreased significantly (P<0.05) in preeclampsia group compared with gestational hypertension group during pregnant metaphase and later stages. The positive expressions and quantity of stained AGEs and RAGEs in placenta tissues increased significantly, and positive expressions of stained MMP-9 decreased significantly in preeclampsia group compared with control group and gestational hypertension group. Conclusion The activation of AGEs-RAGEs system may be closely correlated to preeclampsia occurrence, and MMP-9 is involved in this pathological.
