The effect of Twf1 gene on apoptosis and NF-κB signaling pathway in cardiac myocytes

Abstract:Objective To investigate the effect of inhibiting of Twf1 gene expression on the proliferation, apoptosis and NF-κB signal of cardiomyocytes induced by high glucose.Methods H9c2 cells are stimulated by high glucose, and H9c2 cells were divided into control group (5.5 mmol/L glucose treated cells), NC group (s25mmol/L glucose stimulates cells after transient transfection of siRNA without interference), high glucose group (25 mmol/L glucose stimulated cells after transient transfection of siRNA interfered with Twf1 expression) and Twf1-siRNA group (25 mmol/L glucose stimulated cells after transient transfection of siRNA interfered with Twf1expression).Western blotting was used to detect protein expression;cell viability was detected by CCK8 protein;flow cytometry was used to detect the apoptosis rate and the content of ROS.Results High glucose can increase the expression of Twf1 in H9c2 cells.The expression of Twf1 in H9c2 cells was significantly decreased after transfection with Twf1-siRNA.The cell viability in high glucose group was lower than that in NC group, apoptosis rate, ROS content and p-IκBα, Bax and TNF.The expression ofα-αwas higher than that of NC group (P<0.05).The cell viability of Twf1-siRNA group was higher than that of high glucose group, apoptosis rate, ROS content and p-IκBα, Bax and TNF.The expression ofα-αwas lower than that of high glucose group, and the difference was statistically significant (P<0.05).Conclusion Inhibition of Twf1 gene expression in H9c2 cells can reverse the decrease of cell viability and increase of cell apoptosis induced by high glucose, which may be related to the increase of intracellular ROS content and the decrease of NF-κB signal.
