Influence of simvastatin on myocardial micro-vessels in rats after coronary micro-embolism and potential mechanism

Abstract:Objective To analyze the influence of simvastatin on myocardial micro-vessels in rats after coronary micro-embolism (CME) and potential mechanism. Methods Male SD rats (n=60) weighted from 230 g to 260 g were chosen. All rats were randomly divided into 4 groups: control group (group A), sham-operation group (group B), CME group (group C) and simvastatin intervention group (group D, each n=15). Group D was intragastrically given simvastatin after established CME model, group C, intragastrically normal saline after established CME model, group B, intragastrically normal saline after only opened chest, and group A, only intragastrically normal saline. All rats were killed after 28 d. The levels of myocardial high-sensitivity C-reactive protein (hs-CRP), nitric oxide (NO) and endothelin-1 (ET-1) were detected. The density of myocardial micro-vessels was detected by using immunohistochemistry technique. Results The results of immunohistochemistry technique showed that the density of myocardial micro-vessels decreased significantly in groups C and D compared with groups A and B (all P<0.05). At the same time, the density of myocardial micro-vessels increased in group D compared with group C (P<0.05). The levels of NO and ET-1 decreased significantly and level of hs-CRP increased significantly in group C compared with groups A and B (all P<0.05). The levels of NO and ET-1 increased significantly in group D compared with group C (all P<0.05). Conclusion Simvastatin can protect myocardial micro-vessels after CME possibly through NO.
