Effect of carvedilol on cytokines in myocardial tissue of mice with viral myocarditis

Abstract:Objective To investigate the effect of carvedilol on cytokines in myocardium of mice with viral myocarditis.Methods 4-week-old BALB / c male mice were randomly divided into control group, myocarditis group and myocarditis + carvedilol treatment group, 25 rats in each group. The surviving mice were sacrificed and the heart specimens were taken. The pathological changes of myocardium were observed by light microscope. The plasma norepinephrine content was measured by high performance liquid chromatography-electrochemical method. Fluorescence quantitative test was used to examine the expression of interleukin-6 (IL-6) mRNA in myocardium. Western blot was used to detect the expression of IL-6 protein in myocardium.Results The levels of HW / BW and plasma norepinephrine in the myocarditis group were significantly higher than those in the control group at the 7th and 14th day after treatment (P<0.05). The ratio of HW / BW and plasma norepinephrine in carvedilol group were significantly lower than those in myocarditis group (P<0.05). There were no abnormal pathological changes in the control group. On the 7th and 14th day after treatment, the myocardium infiltration and myocardial degeneration and necrosis of the carvedilol group were significantly less than those in the myocarditis group, and the tissue score was lower than that in the myocarditis group (P<0.05). The expression of IL-6 mRNA and protein in the myocardium of the myocarditis group was significantly higher than that of the control group (P<0.05). The expression of IL-6 mRNA and protein in myocardial tissue of carvedilol group was significantly lower than that in myocarditis group (P<0.05). There was no significant difference in the expression of IL-6 mRNA and protein in the myocardium of the three groups (P>0.05) at 14 days after treatment.Conclusion Carvedilol can inhibit the expression of IL-6 in mice with viral myocarditis and protect the myocardium, which has a good effect.
