Influence of slowly pumping reteplase on ischemic-reperfusion injury in rabbits with acute pulmonary embolism

Abstract:Objective To observe the influence of slowly pumping reteplase and intravenous injecting reteplase on the prognosis of ischemic-reperfusion injury in rabbits with acute pulmonary embolism (PE).Methods Adult health male New Zealand white rabbits of the same age (n=22) were randomly divided into test group and control group, and PE model was established by using autologous thrombus transfusion. After 1 h, the rabbits were given thrombolysis therapy, and test group was given slowly pumping reteplase and control group, rapidly intravenous injecting reteplase. The levels of glutathione peroxidease (GSH-PX), superoxide dismutase (SOD), reactive oxygen species (ROS) and malondialdehyde (MDA) were detected and compared at the time points of 0 h, 1 h, 2 h, 3 h, 6 h and 24 h.Results The levels of ROS and MDA reached peak value, and levels of GSH-PX and SOD reached valley value in control group at the time point of 1 h. The levels of ROS and MDA reached peak value in test group at the time point of 3 h, which was significantly lower than those in control group (P<0.05). The levels of GSH-PX and SOD reached valley value in control group at the time point of 3 h, which had no statistical difference compared with control group (P>0.05). The level changes of ROS and MDA had no statistical difference compared with control group at the time point of 24 h (P>0.05).Conclusion Slowly pumping reteplase delays the oxidative stress peak induced by ischemic-reperfusion injury, and leaves sufficient reacting time to the start of peripheral enzymes for eliminating oxygen free radicals, which is a safe thrombolysis therapy.
