Expression of Profilin-1 protein in aortic tissue and correlation among it and cell injury, apoptosis and ERK1/2 signaling pathway in rats with acute myocardial infarction

Abstract:Objective To investigate the expression of Profilin-1 protein in aortic tissue and correlation among it and cell injury, apoptosis and ERK1/2 signaling pathway in rats with acute myocardial infarction (AMI). Methods Adult male Wistar rats (n=20) were randomly divided into model group and control group (eachn=10), and AMI model was established. The expressions of Profilin-1 protein in aortic tissue and extracellular pERK1/2 protein were detected by using Western blotting, and myocardial endothelial microparticles (EMP) were detected by using flow cytometry. The levels of nitric oxide (NO), cardiac troponin T (cTnT) and creatine kinase MB (CK-MB) were detected by using fluorospectrophotometry. The expressions of P53 mRNA, Fas mRNA, Bax mRNA and Bcl-2 mRNA were detected by using reverse transcription-polymerase chain reaction (RT-PCR).Results The expressions of Profilin-1 and pERK1/2 were higher in AMI group than those in control group (P<0.05). In model group, EMP was higher and NO was lower (allP<0.05). The expressions of cTnT, CK-MB, P53 mRNA, Fas mRNA and Bax mRNA were higher and expression of Bcl-2 mRNA was lower in model group (allP<0.05). The expressions of Profilin-1 and pERK1/2 were positively correlated to cTnT, CK-EMP, P53, Fas and Bax, and negatively correlated to NO and Bcl-2 in model group (allP<0.05).Conclusion The expression of Profilin-1 protein increases in rat model of AMI.
