Expression changes and effective mechanism of endothelial monocyte-activating polypeptideⅡ and C-X-C chemokine receptor type 3 in hyperglycemia-mediated vascular endothelial cell injury

Abstract:Objective To study the expression changes and potential mechanism of endothelial monocyte-activating polypeptide Ⅱ (EMAPⅡ) and C-X-C chemokine receptor type 3 (CXCR3) in hyperglycemia-mediated vascular endothelial cell injury.Methods The vascular endothelial cells were treated with high-level glucose in different doses for 72 h for establishing the model of vascular endothelial cell injury of imitated diabetic retinopathy (DR), and divided into control group and other 3 hyperglycemia groups. The influence of different-dose glucose on cytoactive was detected by using cell counting kit-8 (CCK-8), relevant indexes of cell injury were detected by using detection kit, and expression changes of EMAPⅡ and CXCR3 mRNA were detected respectively by using quantitative polymerase chain reaction (qPCR) and Western blot method.Results The results of CCK-8 detection showed that cell absorbance had statistical difference in 3 hyperglycemia groups compared with control group (P<0.05). The cytoactive decreased significantly in 3 hyperglycemia groups compared with control group, and decreased gradually as the increase of glucose level. The detection results of inflammatory factors and peroxides showed that high-glucose environment induced the release of a large number of inflammatory mediators such as tumor necrosis factor-α (TNF-α), nitric oxide (NO) and reactive oxygen species (ROS) by vascular endothelial cells (P<0.05). The results of Western blot and qPCR showed that the expressions of EMAPⅡ and CXCR3 were significantly up-regulated by high-glucose environment (P<0.05).Conclusion The high-glucose treatment can up-regulate the expressions of EMAPⅡ and CXCR3 in vascular endothelial cells, and further induce the release of inflammatory mediators including TNF-α, NO and ROS and cause cell injury.
