The expression of miR-195 in heart failure and its effect on proliferation of cardiac muscle cells

Abstract:Objective To investigate the expression of microRNA-195 (miR-195) in heart failure and the effects of miR-195 on the proliferation of H9C2 cells.Methods The expression of miR-195 of 56 heart failure patients and 40 normal serum specimens from March 2015 to July 2015 in Shanghai general hospital was detected by Taqman Real-time PCR. The miR-195 mimics was transfected into H9C2 cell by using Lipofectamine 2000. The ability of cell proliferation was detected by MTT and EdU assay. We compared the expression of miR-195 in serum and transfected cardiac muscle cells in patients with heart failure and normal subjects, analyzed of the effects of miR-195 on the proliferation and expression of CDCA4 in cardiac muscle cells.Results ThemiR-195 was up-regulated in serum of heart failure patients compared with normal people serum (P<0.05).MTT results showed that at 48, 72 and 96 hours after transfection, the absorbance of mimic miR-195 group was significantly lower than that of the negative control group (P<0.05). Result of EdU assay showed that cell proliferation in transfected group was inhibited (P<0.05). Result of Western blot results showed that miR-195 inhibited the expression of CDCA4 protein in cardiac muscle cells.Conclusion The miR-195 have a higher expression level in serum of heart failure patients, and it could inhibit the proliferation of H9C2 cells by inhibiting the expression of CDCA4.
