Mechanism of tanshinoneⅡA in inhibiting proliferation of vascular smooth muscle cells

Abstract:Objective To discuss the mechanism of tanshinone ⅡA (Tan ⅡA) in inhibiting the proliferation of vascular smooth muscle cells (VSMC).Methods The abdominal aortic VSMC were cultured by using tissue explant method, and then VSMC were divided into control group [in nutrient solution of 30% fetal bovine serum (FBS, 50 μl)], group A [in nutrient solution of Tan ⅡA (10 mg/L, 25 μl) and 30% FBS (25 μl)], and group B [in nutrient solution of Tan ⅡA (20 mg/L, 25 μl) and 30% FBS (25 μl)]. The cytotoxicity of VSMC was detected by using methyl thiazolyl tetrazolium test (MTT), apoptosis of VSMC was detected by using flow cytometer, and relative expression of cyclin (p21) mRNA was measured by using reverse transcription-polymerase chain reaction (RT-PCR).Results The results of MTT detection showed that the absorbance value was significantly higher in control group than that in group A and group B (allP<0.05), and was significantly lower in group B than that in group A (P<0.05). The apoptosis of VSMC was significantly higher in group A and group B than that in control group, and was significantly higher in group B than that in group A (allP<0.05). Compared with control group, the relative expression of p21 mRNA increased significantly in group A and group B, and was significantly higher in group B than that in group A (allP<0.05).Conclusion Tan ⅡA has significant inhibitory effect on the proliferation of VSMC, and the mechanism maybe related to higher expression of p21.
