Changes of Neuropeptide Y (NPY) and leptin in rats with diabetes complicated with hypertension

Abstract:Objective To discuss the changes of serum and hypothalamus Neuropeptide Y (NPY) and leptin in rats with diabetes complicated with hypertension.Methods SD rats (n=30) were randomly divided into control group, diabetes group and diabetes+hypertension group, and model of type 2 diabetes mellitus and model of diabetes+hypertension were established. The levels of fasting plasma glucose, serum NPY were detected, and expressions of hypothalamus leptin and NPY were detected by using immunohistochemistry technique. Results Compared with control group, the levels of FPG, leptin and NPY increased in diabetes group and diabetes+hypertension group (allP<0.05). The levels of leptin and NPY were higher in diabetes+hypertension group than those in diabetes group and control group [(0.73±0.24) μg/Lvs. (0.46±0.14) μg/L], [(0.73±0.24) μg/Lvs. (0.24±0.05) μg/L], [(300.53±35.74) ng/Lvs. (247.46±40.36) ng/L], [(300.53±35.74) ng/Lvs. (133.52±16.58) ng/L, allP<0.05]. The expressions of leptin and NPY were higher in diabetes group and diabetes+hypertension group than those in control group (allP<0.05). At the same time, the levels of hypothalamus leptin and NPY were higher in diabetes+hypertension group than those in diabetes group (allP<0.05). The level of serum leptin was positively correlated to level of serum NPY (r=0.952,P<0.05), and expression of hypothalamus leptin was positively correlated to expression of hypothalamus NPY (r=0.956,P<0.05) in diabetes+hypertension group.Conclusion The levels of serum and hypothalamus leptin and NPY increase and metabolic disorder is severer in rats with diabetes complicated with hypertension.
