Protective effect of cinnamic acid preconditioning in the myocardial ischemia reperfusion injury of rats by activating Akt signal pathway

Abstract:Objective To explore the effect of cinnamic acid preconditioning on Akt signal pathway and in myocardial ischemia reperfusion injury of rats.Methods 30 male SD rats were randomly divided into three groups:sham operation group (Thoracotomy was accomplished with no anterior descending branch ligation), ischemia reperfusion group, and drug preconditioning group. Myocardial ischemia reperfusion injury model was established by ligating the left anterior descending coronary artery. The apoptosis index of cardiomyocyte was observed by TNUEL (terminal- deoxynucleotidyl-transferase - mediated dUTP-biotin nick end labeling). Western blot technology was used to explore the effects of cinnamic acid preconditioning on Akt signal pathway.Results Compared with ischemia reperfusion group, apoptosis index of drug preconditioning group was significantly decreased (P<0.05). Cinnamic acid pretreatment could increase expression of the phosphorylation Akt (p-Akt) and BCL-2 protein, but had no effect on non-phosphorylation Akt.Conclusion Cinnamic acid preconditioning could reduce apoptosis of cardiomyocyte in myocardial ischemia reperfusion injury model of SD rats. It may induce phosphorylation of Akt, increase expression of BCL-2 and preventing cardiomyocyte apoptosis.
