Effect of telmisartan and amlodipine on myocardial fibroblasts proliferation of neonatal rats induced by angiotensinⅡ

Abstract:Objective To investigate the combination impact of telmisartan and amlodipine on proliferation of neonatal rat cardiac fibroblasts induced by angiotensin Ⅱ (Ang Ⅱ). Methods Primary cardiac fibroblasts were isolated from 30 Wistar neonatal rats and continuously cultured. Cell cultures were divided into five groups and treated with different drugs: control group (cultured with DEME plus 15% fetal bovine serum), Ang Ⅱ group (control medium supplemented with 1×10-6 mol/L Ang Ⅱ), telmisartan group (TE group, addition of 10 μmol/L telmisartan on the basis of Ang Ⅱ group), amlodipine group (AM group, addition of 1 μmol/L amlodipine on the basis of Ang II group), and telmisartan + amlodipine group (TE + AM group, addition of 10 μmol/L telmisartan and 1 μmol/L amlodipine on the basis of Ang Ⅱ group). The cell proliferation of cardiac fibroblasts was measured by CCK8 method. The protein level of lectin-like oxidized low-density lipoprotein receptor-1 (LOX-1) and tumor necrosis factor α (TNF-α) was determined by immunohistochemical staining. The mRNA level of LOX-1 and TNF-α was detected by RT-PCR. Results Cardiac fibroblasts were large and spindle with transparent cytoplasm including 2-3 nucleuses in elliptical shape. The purity was 98%. Compared with the control group, the protein level and mRNA level of LOX-1 and TNF-α were significantly increased in the Ang Ⅱ group (P<0.05). Compared with the Ang II group, the protein level and mRNA level of LOX-1 and TNF-α were significantly reduced in telmisartan group and amlodipine group (P<0.05). Compared with telmisartan group and amlodipine group, the protein level and mRNA level of LOX-1 and TNF-α were further reduced in the combination drug group (P<0.05). Conclusion The induced proliferation of cardiac fibroblasts by Ang II could be inhibited by telmisartan and amlodipine. The combination uses of two drugs yielded more obvious reduction. The possible mechanism might be due to inhibiting the over-expression of LOX-1 and TNF-α.
