Influence of cinnamic acid preconditioning on myocardial ischemia reperfusion injury and possible mechanism in rats

Abstract:Objective To study the influence of cinnamic acid preconditioning on myocardial ischemia reperfusion injury and possible mechanism in rats.Methods Male SD rats (n=34) were divided into sham-operation group (n=10), ischemia reperfusion group (n=12) and preconditioning group (n=12). The model of ischemia reperfusion injury was established through ligation of left anterior descending branch in rats. The sham-operation group was given only thoracotomy without ligation, ischemia reperfusion group was given re-reperfusion for 2 h after ligation for 30 min and preconditioning group was given orally cinnamic acid before the procedure, and the other 2 groups were given orally saline water in equal volume. The area of myocardial infarction (MI) was detected in 3 groups. The changes of cardiomyocytes were observed by using transmission electron microscope (TEM), and expression of extracellular protein regulated kinases1/2 (ERK1/2) signaling pathway was observed by using Western blot technique.Results The area of MI was significantly decreased in preconditioning group (0.18 ±0.05) compared with ischemia reperfusion group (0.34±0.04,P<0.01). The observation of TEM showed that cardiomyocytes were normal in sham-operation group, there were apoptotic bodies observed in ischemia reperfusion group, and there no significant apoptotic phenomenon observed in preconditioning group. The phosphorylation of ERK1/2 (p-ERK1/2) was induced by cinnamic acid preconditioning, and expression of p-ERK1/2 was significantly higher in preconditioning group than that in ischemia reperfusion group (P<0.01). The expression of Bax (the downstream protein of ERK1/2) decreased in preconditioning group compared with ischemia reperfusion group (P<0.01). The cinnamic acid preconditioning had no significant influence on the expression of total ERK1/2 (non-p-ERK1/2).Conclusion The cinnamic acid preconditioning can relieve myocardial ischemia reperfusion injury, and the mechanism maybe related to the phosphorylation of ERK1/2, decrease of Bax expression, inhibition of apoptosis and protection of myocardium.
