Effect of severe maternal hyperglycemia on sugar and fat metabolism and atherosclerosis in Sprague-Dawley rat offspring

Abstract:Objective By intraperitoneal injection of streptozocin(STZ)on maternal, to investigate the effect of maternal hyperglycemia on sugar and fat metabolism and atherosclerosis in SD (Sprague-Dawley) rat offspring.Methods SD rats (8-week-old, weighing 200~250 g) were used in this study. The maternal control group, diabetes group were established according to the experimental requirements. Maternal diabetes group was induced by a 40 mg/kg intraperitoneal injection of STZ on the first day of gestation. Maternal control group was injected with citrate buffer. The offspring were feeding normally and marked as offspring control group and offspring diabetes group. Glucose and insulin tolerance tests (GTT and ITT) were performed; and serum levels of lipids were measured in offspring groups. PCR was performed to analyze mRNAs expression of E-selectin, VCAM-1, ICAM-1 and ICAM-2 in these groups.Results Given 40 mg / kg STZ to pregnant rats can lead to gestational diabetes. Compared with offspring control group, offspring diabetes group showed more severe impairment in GTT and ITT,had higher level of blood lipid and mRNAs expression of E-selectin, VCAM-1, ICAM-1.Conclusion Severe maternal hyperglycemia may be a high-risk factor of sugar and fat metabolism and atherosclerosis in offspring.
