Regulation effect of atorvastatin on PPARγ in peripheral monocytes in patients with acute coronary ;syndrome

Abstract:Objective To observe the regulation effect of atorvastatin on PPARγin peripheral monocytes in patients with acute coronary syndrome (ACS). Methods ACS patients (n=124) were chosen as ACS group and normal persons (n=35) were chosen as control group from Jul. 2011 to Dec. 2012. ACS group was given atorvastatin on the base of routine drugs, and then divided randomly into low-dose group (10 mg/d) and high-dose group (40 mg/d, each n=62). On the second day after hospitalization and after atorvastatin intervention for 1 w, the changes of biochemical indexes of blood fat and hsCRP were detected conventionally, and the expression of PPARγprotein in peripheral monocytes was detected by using flow cytometry and fluorescence immunoassay. Results The expression of PPARγprotein was observed in control group and ACS group, and level of hsCRP was significantly higher in ACS group (12.07±2.01 mg/L) than that in control group (2.99±0.15 mg/L, P<0.01), and expression of PPARγprotein was lower in ACS group (2.35±0.23) than that in control group (5.93±0.54, P<0.05). After atorvastatin intervention for 1 w, the level of hsCRP decreased slightly, and expression of PPARγprotein increased slightly in low-dose group but the difference was not statistically significant before and after treatment (P>0.05). The level of hsCRP decreased significantly and expression of PPARγprotein increased significantly in high-dose group and the difference was statistically significant before and after treatment (P<0.01). The descent range of level of hsCRP (-11.45±2.14) and ascent range of expression of PPARγprotein (+3.38±0.12) in high-dose group before and after treatment were more significant than those in low-dose group (P<0.01). Conclusion The expression of PPARγprotein decreased significantly in ACS patients, and atorvastatin can ameliorate the expression showing a dose-independent manner.
