Artificial intelligence reshaping lipid nanoparticle-mediated nucleic acid delivery

Abstract:Harnessing its powerful capabilities in data processing and intelligent decision-making,artificial intelligence(AI)has shown significant advantages and promising potential in advancing the development of nucleic acid delivery systems.This article provides a systematic review of how AI technologies are reshaping the research and development of lipid nanoparticles(LNPs),the leading nucleic acid delivery platform,detailing recent progress and breakthroughs in molecular design,targeted delivery,and prediction of in vitro and in vivo performance.Strategies for AI-enabled clinical translation are highlighted,including multimodal data-guided tissue-specific delivery,closed-loop"design-experiment-feedback"optimization via automation,and digital twin-based cross-species prediction.The article also examines current challenges in AI-driven LNP development,such as data quality and model interpretability,and looks forward to next-generation AI technologies that integrate causal inference and physics-informed modeling to achieve"designable,predictable,and verifiable"LNPs,offering valuable insights for nucleic acid drug delivery.
