Advances in nanodrug delivery systems for acute myeloid leukemia therapy:novel strategies for synergistic efficacy,targeted delivery and drug resistance overcoming

Abstract:Acute myeloid leukemia(AML)is a highly heterogeneous hematologic malignancy with high mortality and poor prognosis.Over the past 45 years,the combination of cytarabine and daunorubicin has remained the primary chemotherapy regimen for AML.However,challenges such as nonspecific drug distribution,chemotherapy-induced toxicity,and high heterogeneity of AML cells result in high rates of treatment failure and relapse.In recent years,rapid advances in sequencing technologies have enabled a deeper understanding of the molecular and cytogenetic characteristics of AML,shifting research toward multi-pathway combination therapies.Nanodrug delivery systems(NDDSs)provide essential technical support for multi-target combination therapies and have become a promising novel strategy in AML treatment.This review summarizes the advantages of NDDSs in AML therapy,highlighting recent advances in synergistic effects,active targeting,and resistance reversal.It also explores the potential of NDDSs to revolutionize AML treatment and proposes new strategies to address key challenges in targeted delivery and drug resistance.
